Zhigancao decoction (100%) has no obvious effect on the heart rate and electrocardiogram of rabbits after intravenous injection of epinephrine hydrochloride 50mg/kg at 10 ml/kg, but it can significantly shorten the duration of arrhythmia and obviously accelerate the recovery time of normal heart rhythm.
2. Affect the physiological characteristics of isolated myocardium. The right atrium of rats with sinoatrial node was suspended in a bath containing 20ml tyramine, and 6.4g/L licorice soup (4g crude drug /ml) was added. After 10 minute, the self-discipline of isolated right atrium was obviously inhibited, and the frequency was from 176.4 13.8 times/minute before administration. 3. The action potential of isolated myocardium is affected. Guinea pig right ventricular papillary muscle was perfused into improved Taylor's solution, and 6.4g crude drug /L roasted licorice soup was added. The action potential duration (APD) and effective refractory period (EPR) of action potential of guinea pig papillary muscle cells were significantly prolonged, and the maximum depolarization rate (Vmax) was decreased. These effects are dose-dependent, but have no obvious effect on resting potential (RP).
4. Anti-myocardial ischemia-reperfusion injury: Ligating the anterior descending coronary artery (LAD) and reperfusion with 4.8g/L Zhigancao Decoction injection can significantly reduce the total incidence of ventricular tachycardia (VT), ventricular fibrillation (VF) and arrhythmia during reperfusion in rats, and inhibit the release of creatine kinase (CK) and lactate dehydrogenase (LDH) in myocardial cells. Zhigancao decoction 2,4,8 g/L can significantly reduce the content of malondialdehyde (MDA) and increase the activity of superoxide dismutase (SOD) in reperfusion myocardium. The above effects of Zhigancao decoction are similar to those of calcium antagonist verapamil (0.5 μ mol/L). Giving rats Zhigancao Decoction (1 1.4g crude drug /kg) daily 1 time for 4 consecutive days can significantly reduce the total incidence of ventricular premature beats and arrhythmia induced by reperfusion. The same dose of Zhigancao decoction can significantly reduce the range of myocardial infarction after reperfusion, and significantly reduce CK activity, LDH activity and MDA content in serum after myocardial ischemia and reperfusion.
5. Experimental model of anti-Yin deficiency rats: Rats were injected with L-thyroxine 0.5mg/kg subcutaneously for 5 days, and the syndrome model of "Yin deficiency" was reproduced. (1) Anti-arrhythmia in rats with yin deficiency. Intravenous injection of 0.5ml/ 100g Radix Glycyrrhizae Preparata can obviously slow down the heart rate, eliminate sinus arrhythmia and reduce the incidence of ventricular premature beats. Intravenous injection of Zhigancao Decoction can significantly improve the arrhythmia caused by intravenous injection of adrenaline, which mainly shows that the occurrence time of ventricular premature beats (VP), ventricular tachycardia (VT), conduction block (AVB) and other arrhythmias is delayed, the incidence rate is reduced, and the dose of adrenaline used to induce arrhythmia is significantly increased. Compared with healthy rats, Radix Glycyrrhizae Preparata injection has a better curative effect on arrhythmia in rats with yin deficiency, and the score of yin deficiency group is 665438 0% lower than that of control group. However, the score of the healthy drug group dropped by 32%. (2) Improvement of Yin deficiency syndrome: Oral administration of 5g/kg Zhigancao Decoction (1g/ml) 10 day in rats with Yin deficiency can obviously reduce the concentration of serum thyrotropin-releasing hormone, but has no obvious effect on T3 and T4 levels; The level of plasma cAMP decreased from 65438±099±65438±02 pmol/ml/ml in the control group to 65438 060 20 pmol/ml in the administration group. 6. Anti-hypoxia Zhigancao decoction can prolong the death time of mice due to decompression and hypoxia asphyxia and reduce the death rate of mice due to hypoxia. When only 5.9% of the control group survived, the survival rate of the people who were given the decoction reached 76.5%.
