(1) Arrangement of research progress

Kawasaki disease is an acute, self-limiting systemic vasculitis that occurs in infants and young children under the age of 5. The disease was first discovered in Japan in 1962, first reported by the Japanese scholar Tomisaku Kawasaki in 1967, and has been reported in Europe, Asia, the United States, Australia, and South Africa since then. Since the etiology of this disease is unknown, the clinical manifestations are complex, and multiple systems of the body can be involved, especially the coronary arteries, in Japan and the United States has replaced rheumatic fever as the most common disease of acquired heart disease in children, and it has become one of the most common acquired heart diseases in children, and it may be one of the risk factors for ischemic heart disease in adults. The diagnosis of Kawasaki disease is confirmed when at least 5 of the above criteria are met, with 1) being a requirement. By examining the effects of chemokine CCL5/ CCR5 on T-cell chemotaxis, our experimental conclusions are as follows:

Binding of the chemokine CCLS to its receptor CCRS promoted motility, invasiveness, chemotaxis, and proliferative effects in peripheral cells of children with Kawasaki disease, the mechanism of which may be that the CCL5/ CCR5 bioaxis initiates calcium-ion-related signaling within the cancer cells by causing alterations in intracellular calcium ion concentrations.

(4) Difficulties and problems

Chemokines, a member of the cytokine superfamily, are a class of small-molecule proteins with a directed chemoattractive effect on cells, which can directionally attract immune cells to participate in the immune response and immunopathological processes [12]. To date, at least 50 chemokines have been identified, which are composed of 70-125 amino acids with molecular weights ranging from 8-11 KD. Most chemokines contain two characteristic disulfide bonds formed by four conserved cysteines, so they have a very similar high-level structure with an α-helix at the C-terminus and an irregular N-terminus, and have a relatively high biological activity to bind to receptors.

Chemokines were first discovered as chemotactic mediators, but with the in-depth study of chemokines, it is now recognized that chemokines not only have chemotactic effects on leukocytes, but also play a wide range of physiological and pathological roles in the development of immune cells and organs, the process of immune response, inflammatory responses, pathogen infections, trauma repair, as well as tumor formation and metastasis, etc. [14-16].
