What is a real-time fluorescence quantitative pcr instrument?

The main features of the Model 7900HT Fluorescence Quantitative PCR Instrument:

●Efficient platform-The 7900HT system is built to support high throughput, and the optical system is optimized for activation of fluorescent signals in 384-well plates and real-time detection.

●Reliable Results-The 7900HT System utilizes the same quantification principles as the 7700 and 5700, utilizing the correspondence between the Ct value (Ct is defined as the PCR cycle that produces a statistically significant and detectable fluorescence emission above the baseline) and the starting copy number, and accurately calculating the starting copy number of unknown samples from an external standard curve.

●Highly Reproducible-The fluorescence detection of the 7900HT System occurs at the beginning of the logarithmic phase of PCR, so there is no limiting factor for each component in the reaction tube, ensuring that the results are accurate and reproducible.

●Advanced Fluorescence Detection Technology - The RISM? 7900HT fluorescence quantitative PCR system effectively increases the yield without decreasing the speed and resolution, which is based on the advanced fluorescence detection technology: a laser beam scans and activates fluorescent dyes in each of the 384 wells, and fluorescence signals are separated by a grating, and the separated multicolored fluorescence arrives at the CCD camera simultaneously, realizing the simultaneous detection of multicolored fluorescence. The CCD camera is able to detect multiple colors of fluorescent light at the same time.

●Autosampling Unit - The 7900HT system is equipped with automated accessories for maximum efficiency. The automation unit can hold up to 84 384-well plates, and the robotic arm transports the plates to be tested to the mainframe in a certain order. The bar code reader automatically recognizes the plate numbers entering the mainframe, allowing you to insert or replace plates in the sample train at any time.

●Variety of chemistries-The chemistries available on the Model 7900HT Fluorescence PCR Instrument include TaqMan fluorescent probes and SYBR Green fluorescent dyes, which are highly sensitive and specific for real-time quantification of multiplexed reactions or single-nucleotide polymorphisms (SNPs).
SYBR Green dyes are used for target identification (screening assays). SYBRGreen dye is ideal for target identification (screening assays) or in assays where only a few reactions are required.

●Effective Contamination Prevention - The 7900HT System utilizes specially designed and always closed sample tubes throughout the PCR process. These tubes are closed immediately after the addition of samples and reagents and remain closed during amplification and detection, which greatly reduces the chance of contamination; in addition, there is no PCR post-processing on the 7900HT System, effectively preventing false positives due to contamination of the experimental environment with amplification products.

●Multiple quantification methods-ABIPRISM? 7900HT Sequence Detection System software provides a quantitative method for gene expression. Absolute quantification is the direct determination of the amount of a target (gene) from a standard curve. Relative quantification methods calculate the amount of an unknown sample by comparing it to a calibrated sample and do not require a standard curve.

1. Absolute Quantification

To determine the amount of a target (gene) in an unknown sample, the 7900HT System can be used to determine the CT of the sample, and then a standard curve is used to determine the starting copy number. In drawing the standard curve, the 7900HT System software first calculates the CT values for several dilutions with known starting copy numbers. Next, the software plots the logarithmic value of the measured CT value corresponding to the starting copy number. The relationship is linear over a range of at least five orders of magnitude.

2. Relative quantification

Relative quantification is ideal for gene expression studies. Instead of applying a standard curve, expression levels are calculated relative to a calibration sample. An endogenous control is used to calibrate the amount of sample. This approach, combined with the ability of the 7900HT system to quantify both the target and the endogenous control in a single multiplex reaction, enables efficient, high-throughput gene expression studies.

Effective SNP Detection

In addition to real-time quantitative assays, the 7900HT System includes software for large-scale screening of known single nucleotide polymorphisms (SNPS). In the one-allele system, a multiplex PCR reaction is performed in the same tube with two TaqMan probes corresponding to the two alleles. The fluorescence generated at the termination of the reaction is measured with the 7900HT system, and the results are rapidly obtained by the analysis software.
